OK so make sure you don't just start prying it off. First go around the side opposite you want to take off first and pop the tire down off of the bead all around. Of course make sure rim lock is loose. Then flip it over and pop it off the bead there. Then take a nice hefty bite and pry it over the rim. Hold that there and then go for another bite. Hold that there and go for another. If you have to go more than three to get the tire to stay on the rim pull the middle tool out. I do this process with one small tire spoon and two big blunt screwdrivers.

The rim lock is a rubber piece on the inside of the rim that has a stud that sticks thru the rim and then uses a nut to hold it to the rim. The stud sticks out of the rim about halfway around the rim from where the schrader valve is.

I don't know a dirt bike that doesn't have one. I have changed tires on bikes all the way back to the 70's and they still had it.
